▎ 摘 要
This work demonstrates a novel approach toward development of advanced immunoassay based on chemically functionalized magnetic graphene sensing platform, and the preparation, characterization, and measurement of relevant properties of the immunosensing probe useful for determination of neuron-specific enolase (NSE) in biological fluid. The magnetic immunosensing probes based on the combination of graphene nanosheets with magnetic nanoparticles show good covalent conjugation properties for polyclonal rabbit anti-human NSE antibody selective to NSE. Magnetic immunosensing probes present good magnetic properties to facilitate and modulate the way while integrating onto a magnetic carbon paste electrode, which exhibiting good electrochemical response for detection of NSE, and allow detection of NSE at the concentration as low as 0.3 ng/mL. Intra- and inter-assay coefficients of variation were below 10.5%. In addition, the methodology was validated by assaying spiking serum samples, and the relative standard deviation values between the potentiometric immunoassay and commercialized ELISA were 2.3-10.9%.
