NOVELTY - Method for preparing sensor involves (a) polishing glassy carbon electrode, soaking electrode in chloroauric acid, electroplating and drying electrode, (b) soaking the electrodes in solution comprising anti-hepatitis B surface (anti-HBs) antibody and anti-hepatitis B envelope (anti-HBe) antibody, adding bovine serum albumin, rinsing electrode and drying electrode, and (c) soaking electrode obtained in step (b) in mixture containing tin dioxide (SnO2)-graphene (GS)/gold (Au)/platinum (Pt)/nanoparticles (NPs)-HBs-antibody (Ab)2 solution and storing electrode in refrigerator. USE - The method is useful for preparing sensor, which is used for simultaneously detecting HBs and HBe marker (all claimed). ADVANTAGE - The method has high specificity and sensitivity for detecting HBs and HBe marker. DETAILED DESCRIPTION - Method for preparing sensor involves (a) polishing glassy carbon electrode using aluminum oxide polishing powder, where the glassy carbon electrode has diameter of 3-5 mm, washing electrode using ultrapure water, soaking electrode in 10 ml of 1 %mass chloroauric acid, electroplating at 0.2 V for 30 seconds, drying electrode at room temperature and rinsing electrode using ultrapure water, (b) soaking the electrodes in 8-12 mu g/ml solution comprising anti-hepatitis B surface (anti-HBs) antibody and anti-hepatitis B envelope (anti-HBe) antibody at 4 degrees C for 12 hours, adding 3 mu l of 1-3 mg/ml of bovine serum albumin for blocking non-specific active site on electrode surface, rinsing electrode, storing electrode at 4 degrees C in refrigerator, soaking electrodes in series of different concentrations of 0.1 pg/ml-50 ng/ml HBs and HBe antibodies at 37 degrees C for 50 minutes, drying electrode at room temperature, rinsing electrode and again drying electrode at 4 degrees C, and (c) soaking electrode obtained in step (b) in 1-3 mg/ml mixture comprising tin dioxide (SnO2)-graphene (GS)/gold (Au)/platinum (Pt)/nanoparticles (NPs)-HBs-antibody (Ab)2 solution and SnO2-GS/silver (Ag)-cysteine-AuNPs-HBe-Ab2 solution at room temperature for 50 minutes, and storing electrode in refrigerator at 4 degrees C. An INDEPENDENT CLAIM is included for method of simultaneously detecting HBs and HBe antigens.
