▎ 摘  要

NOVELTY - Assay kit comprises a silicon dioxide-based surface construct comprising a capture molecule bound to the surface of the silicon dioxide-based surface construct, where the capture molecule is capable of binding to the target biomolecule, a nanozyme comprising a probing molecule bound to the nanozyme, where the probing molecule is capable of binding to the target biomolecule, and a visualization agent, where the visualization agent comprises a detecting molecule, and the detecting molecule is capable of binding to the nanozyme and is configured to change color upon a chemical reaction with the nanozyme. USE - The assay kit is used for detecting target biomolecule in sample of bodily fluid of subject. The target biomolecule is chosen from SARS-CoV-2 antibodies, including SARS-CoV-2 variant neutralizing antibodies, such as δ variant SARS-CoV-2 neutralizing antibodies, M proteins and non-M proteins of Streptococcus group A, anti Streptococcus A antibodies, influenza virus A and B antigens, anti influenza A/B antibodies, HIV p24 capsid protein, anti-HIV antibodies, chlamydial LPS, anti-Chlamydia trachomatis antibodies, norovirus antigens, anti norovirus antibodies, rotavirus antigens, anti rotavirus antibodies, flavivirus antigens and zika virus antigens (all claimed). ADVANTAGE - The assay kit provides convenient, rapid, reliable and low-cost diagnostic tests for viral detection, and provides nanozyme which converts the detecting molecule into colored form upon interaction without need to use any additional reagent, which significantly reduces an analysis time. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for the following: a method for producing a glass surface construct for the assay kit for detecting a target biomolecule in a sample of bodily fluid of a subject, involving (i-1) providing a glass surface, (ii-1) contacting the glass surface with an activating agent to produce an activated glass surface, (iii-1) contacting the activated glass surface with an active polymer to produce a modified glass surface, (iv-1) contacting the modified glass surface with a capture molecule, (v-1) Optionally, contacting the modified glass surface of step (iv) with a surface blocking agent, preferably bovine serum albumin (BSA) or casein; and a method for detecting a target biomolecule in a sample of a bodily fluid of a subject, involving (i-2) providing the assay kit, (ii-2) providing the sample of a bodily fluid of a subject, (iii-2) contacting the silicon dioxide-based surface construct with the nanozyme and the sample of a bodily fluid for at least 2 minutes, preferably 15 minutes, (iv-2) separating the silicon dioxide-based surface construct from the mixture of step (iii), (v-2) optionally, washing the separated silicon dioxide-based construct of step (iv) with a cleaning buffer, and (vi-2) contacting the silicon dioxide-based construct of step (v) with the visualization agent for at least 10 seconds, preferably 2 minutes.
