NOVELTY - A functionalized solid-phase micro-extraction probe is prepared by coating a graphene oxide-polyacrylonitrile solution on an activated stainless-steel needle and polymerizing a norepinephrine solution on its surface to form a polynorepinephrine (PNE) coating. The graphene oxide-polyacrylonitrile solution is prepared by mixing polyacrylonitrile solid and N,N-dimethylformamide, stirring to form a viscous suspension, heating to 80-100°C to form a yellow clear and transparent polyacrylonitrile glue, cooling to room temperature, adding graphene oxide powder to N,N-dimethylformamide, ultrasonically dispersing to obtain graphene oxide solution, adding to the polyacrylonitrile glue and uniformly stirring. The activated stainless-steel needles are obtained by cutting medical stainless steel needles with a diameter of 0.4-0.6 mm into long segments, ultrasonically pretreating the segment in concentrated hydrochloric acid for 8-12 minutes, removing and washing with deionized water. USE - Functionalized solid-phase micro-extraction probe used in detection of toxin content in living puffer fish (claimed) for safe eating of fish. ADVANTAGE - The functionalized solid-phase micro-extraction probe is prepared by simple preparation process, and has high sensitivity and detection limit, which can meet the needs of puffer fish for safe consumption. The detection method is short and economical, and can be completed within 1 hour, and does not need to kill the puffer fish for detection and effectively avoids the problem of sample lethality caused by the detection of puffer fish toxin. DETAILED DESCRIPTION - A functionalized solid-phase micro-extraction probe is prepared by coating a graphene oxide-polyacrylonitrile solution on an activated stainless-steel needle and polymerizing a norepinephrine solution on its surface to form a polynorepinephrine (PNE) coating. The graphene oxide-polyacrylonitrile solution is prepared by mixing polyacrylonitrile solid and N,N-dimethylformamide in mass ratio of 1:7-8, stirring to form a viscous suspension, heating to 80-100°C, heat preserving for 50-70 minutes to completely dissolve the polyacrylonitrile to form a yellow clear and transparent polyacrylonitrile glue, cooling to room temperature, adding graphene oxide powder to N,N-dimethylformamide in mass-to-volume ratio of 15-25 mg: 200-300 μL, ultrasonically dispersing to obtain graphene oxide solution, adding to the polyacrylonitrile glue and uniformly stirring. The activated stainless-steel needles are obtained by cutting medical stainless steel needles with a diameter of 0.4-0.6 mm into 2-3 cm long segments, activating surface by ultrasonically retreating the segment in concentrated hydrochloric acid for 8-12 minutes, removing and washing with deionized water. An INDEPENDENT CLAIM is included for use of the functionalized solid-phase micro-extraction probe in detection of puffer fish toxin in vivo.
