NOVELTY - Preparing polymeric composite involves modifying gold electrode with reduced graphene oxide nanocomposite followed by electrochemical polymerizing poly (3-hydroxyphenylacetic acid) polymer for incorporation of oligonucleotide probes. Immobilization is performed by adsorption of oligonucleotide probe sequence comprises 16 nucleobase, that is 5'-GGGGGGGGAAAAAAAA-3'. The complementary target is detected directly in real time using the differential pulse voltammetry technique. USE - Method for preparing polymeric composite. ADVANTAGE - The method enables to provide wide applicability, sensitivity, low cost, reduced sample volume and real-time analysis. DETAILED DESCRIPTION - Preparing polymeric composite based on reduced graphene oxide and poly (3-hydroxyphenylacetic acid), as matrix for oligonucleotide hybridization, involves modifying gold electrode with reduced graphene oxide nanocomposite followed by electrochemical polymerizing poly (3-hydroxyphenylacetic acid) polymer for incorporation of oligonucleotide probes. Immobilization is performed by adsorption of oligonucleotide probe sequence comprises 16 nucleobase, that is 5'-GGGGGGGGAAAAAAAA-3'. The complementary target is detected directly in real time using the differential pulse voltammetry technique. The complementary target is detected in indirect way in real time using signal amplifier by indicator of formation of double DNA tape like ethidium bromide, monitoring the signal by the technique of differential pulse voltammetry.
