NOVELTY - Rectal gel is prepared by preparing hydroxybutyl chitosan by a homogeneous synthesis method and graphene oxide by an improved Hummers method, carrying an anticancer model medicine X on the surface of graphene oxide to obtain graphene oxide-X, and finally cross-linking graphene oxide-X with hydroxybutyl chitosan to obtain hydroxybutyl chitosan/graphene oxide-X. USE - The rectal gel is useful as rectal medicinal carrier (claimed). ADVANTAGE - The rectal gel improves deficiency of in situ temperature sensitive gel drug loading and its mechanical strength, and drug release amount and bioavailability. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for method for preparing the rectal gel, which involves (a) adding potassium hydroxide, urea, distilled water, and chitosan to the container in sequence, mixing and refrigerating the mixture for a certain period of time, maintaining to room temperature, adding 1,2-butylene oxide to the mixture, reacting the mixture under nitrogen protection, adjusting the pH, dialyzing and freeze-drying to obtain hydroxybutyl chitosan, (b) adding concentrated sulfuric acid, graphite powder, sodium nitrate and potassium permanganate into container, finishing the reaction after reacting for a period of time at a certain temperature, adding hydrogen peroxide solution until the color of the reaction solution turns yellow, centrifuging the product, washing the product, and freeze-drying the product to obtain graphene oxide, (c) preparing a certain amount of graphene oxide dispersion liquid by ultrasonic stripping method, adding the medicine X aqueous solution, reacting the solution in shading, centrifuging the product after the reaction and freeze-drying the lower layer to obtain graphene oxide-X, and (d) adding hydroxybutyl chitosan and Bletilla striatapolysaccharide to the graphene oxide-X and stopping reaction after a period of time.
