▎ 摘 要
NOVELTY - Preparing a graphene oxide-DNA sensor based on an enzymatic cycle amplification comprises e.g. preparing graphene oxide through the improved Hummers method, vacuum drying, carrying out ultrasonic dispersing in the aqueous solution to obtain the graphene oxide dispersion, activating graphene oxide containing 50Mm NHS and 200mM EDC aqueous solution A, using 2 mg/ml graphene oxide dispersion, adding super-pure water, reacting at room temperature for 0.5 hours and eluting and storing. USE - The sensor is useful for detecting thrombin (claimed). ADVANTAGE - The sensor increases detection limit of thrombin to 0.024 pM. DETAILED DESCRIPTION - Preparing a graphene oxide-DNA sensor based on an enzymatic cycle amplification comprises (i) preparing graphene oxide through the improved Hummers method, vacuum drying, carrying out ultrasonic dispersing in the aqueous solution to obtain the graphene oxide dispersion, (ii) activating graphene oxide containing 50Mm NHS and 200mM EDC aqueous solution A obtained in step (i) 2 mg/ml graphene oxide dispersion, adding super-pure water, reacting at room temperature for 0.5 hours and eluting, storing, (iii) preparing GO-aptamer sensor, GO surface joint capture DNA aminated prepared in the step (iii), dripping TBA, binding with the capture DNA, adding polyethylene glycol, to obtain the GO-DNA sensor. INDEPENDENT CLAIMS are also included for preparing graphene oxide-DNA sensor by imprinted polymer; and detecting thrombin time comprising (i) detecting the fluorescence value of the GO-DNA sensor, (ii) adding thrombin in GO-DNA sensor, adding exonuclease exonuclease, or directly adding the mixture of thrombinexonuclease Thrombin-Exonuclease. after reacting for 30 minutes, detecting the fluorescent value and analyzing the change in fluorescence.
