NOVELTY - Preparation of calmodulin-specific binding target peptide-modified reduced graphene oxide nanoparticle comprises sonicating 0.5-5 mg/mL graphene oxide (GO) dispersion for 30-120 minutes, centrifuging to obtain micron-sized GO, filtering through microporous membrane to obtain nano-GO, binding GO and calmodulin to target peptide at 50-80 degrees C, adding potassium hydroxide and water at mass ratio of 20-50:2-5:100-500:2000-10000, and stirring for 10-20 hours; and cooling to room temperature, and eluting through ultrafiltration tube. USE - Method for preparation of calmodulin-specific binding target peptide-modified reduced graphene oxide nanoparticle used for regulating ion channels or in-vitro regulating of calmodulin conformational changes (all claimed). ADVANTAGE - The method is capable of remote control process. The product is suitable for treatment of diseases related to ion channels, such as atrial fibrillation. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for regulating ion channels comprising inoculating cells in 24-well plate plated with cell slides, growing for 24 hours, adding 50 mu L incubated liquid to each well, culturing for 18-36 hours, adding calmodulin-specific binding target peptide-modified reduced graphene oxide nanoparticle, placing to incubator, and incubating for 2-6 hours to obtain 5-50 g/mL cells; and removing climbing slide, washing with buffer, and irradiating cells on climbing plate with near-infrared light for 3-8 minutes. The incubating comprises mixing 2 g ion channel plasmid with 6 L X-tremeGENEHP DNA transfection reagent in 200 L Opti-MEM medium, and standing for 20 minutes.
