NOVELTY - Method of identifying a protein biomarker from a biological fluid, involves (a) incubating a nanoparticles with a biological fluid sample from a subject in a disease state to allow biomolecule coronas to form on the surface of the nanoparticles, (b) isolating the nanoparticles and surface-bound biomolecule coronas, and (c) analyzing the biomolecule coronas by mass spectrometry to identify a previously unknown disease-specific protein biomarker, where the analysis is performed at least in portion using a computer, or a method of identifying a protein biomarker from a biological fluid, involves incubating a nanoparticles that differ from each other in at least one physicochemical property with a biological fluid sample from a subject in a disease state to allow biomolecule coronas to form on the surface of the nanoparticles, isolating the nanoparticles and surface-bound coronas, and analyzing the coronas by mass spectrometry to identify the biomarker associated with the disease state. USE - The method is useful for identifying protein biomarker from biological fluid, and detecting diseases including cancer such as lung cancer, melanoma or ovarian cancer (all claimed), or disorder such as endocrine disorder, cardiovascular disease such as coronary artery disease, inflammatory disease or neurological disease such as Alzheimer's disease. ADVANTAGE - The method improves our ability to detect cancer at very early stages, and is accurate, sensitive and predictive.
