NOVELTY - A modified gold electrode comprises graphene oxide, gold nanotube, locked nucleic acid probe and signal amplifying probe composition, where the gold electrode surface is modified with graphene oxide and zinc oxide sol-gel composition, locked nucleic acid probe and blocked non-specific adsorption sites. The locked nucleic acid probe recognizes New Delhi metallo- beta -lactamase-1(NDM-1)gene specifically. USE - Modified gold electrode for use in an electrochemical biosensor that is utilized for detecting NDM-1 gene (all claimed). ADVANTAGE - The electrochemical biosensor has wide market value, and detects NDM-1 gene rapidly with high specificity and sensitivity and detects multi-drug resistant bacteria with simple, rapid, sensitive and specific tool. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are also included for the following: (1) a method for producing a modified gold electrode, involves preparing graphene oxide and zinc oxide sol-gel by dissolving graphene oxide in water, and mixing obtained suspension with zinc oxide sol-gel and anhydrous ethyl alcohol uniformly to obtain graphene oxide and zinc oxide sol-gel, preprocessing gold electrode by grinding, polishing, cleaning and drying the gold electrode, modifying the gold electrode by coating the graphene oxide and zinc oxide sol-gel on the preprocessed gold electrode, drying the gold electrode, immersing the gold electrode in gold nanotube solution, cleaning the gold electrode, immersing the gold electrode in solution-containing locked nucleic acid probe, cleaning the gold electrode, immersing the gold electrode in mercaptohexanol solution blocked for non-specific absorbing sites to obtain modified gold electrode, and preparing signal amplification probe by adding locked nucleic acid probe to gold nanotube solution, oscillating and blending the mixture uniformly, reacting the mixture completely, centrifuging the mixture, collecting precipitate, washing the precipitate with phosphate buffered saline (PBS) to obtain signal amplifying probe in PBS liquid, and storing the product (2) an electrochemical biosensor comprising working electrode, counter electrode, reference electrode and test substrate liquid, where the working electrode is modified gold electrode, counter electrode is molybdenum electrode, reference electrode is saturated calomel electrode, and the test substrate liquid has pH of 7.4 and comprises 2 mmol/l potassium ferricyanide-potassium ferrocyanide and 5 mmol potassium chloride in PBS solution.
