NOVELTY - Visually detecting Salmonella gene by single-labeled single-stranded (ss)DNA probe comprises (i) extracting the genomic DNA of the sample to be tested, (ii) expressing and purifying clustered regularly interspaced short palindromic repeats (CRISPR) associated protein 12a (Cas12a), (iii) designing and synthesizing recombinase polymerase amplification (RPA) primers and CRISPR RNA (crRNA), (iv) performing RPA amplification including using the genomic DNA extracted in step (i) as a template, amplifying in an RPA system containing RPA primers to obtain an RPA product, (v) performing fluorescence visual detection including uniformly mixing the RPA product obtained in the step (iv), Cas12a obtained in the step (ii), crRNA obtained in the step (ii), single fluorophore-labeled ssDNA probe and NEBuffer2.1 buffer solution, incubating for 10-60 minutes, adding graphene oxide (GO), and judging whether the sample to be tested contains Salmonella gene by the fluorescence signal. USE - The method is useful for visually detecting Salmonella gene by single-labeled ssDNA probe. ADVANTAGE - The method: realizes the specific, single marker and visual detection of Salmonella gene; and converts the genetic information of Salmonella into visible fluorescence changes.
