NOVELTY - Preparing immunochromatographic test strip marker graphene-colloidal gold involves mixing graphene powder and pyrene formic acid by weight 1-2:0.5-1.5, adding in methanol, fully reacting 1-8 hours (h) to obtain carboxylated graphene, centrifuging the carboxylated graphene, after cleaning and drying, obtain the carboxylated graphene powder, adding 30-70mM 2-(N-morpholino)ethanesulfonic acid (MES) solution to it, after 5-20 minutes (min), dropping 1-3mM 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) and 4-6mM sulfo- N-hydroxysuccinimide (sNHS) reaction, stirring under the condition of 50-100rpm for 10-40min to obtain aminated graphene, and then centrifuging for 10-40min under the condition of 5000-15000 revolutions per minute (rpm). The precipitate is resuspended, and is prepared into a 0.1-0.3mg/ml aminated graphene solution. USE - Method for preparing immunochromatographic test strip marker graphene-colloidal gold. ADVANTAGE - The method enables to prepare immunochromatographic test strip marker graphene-colloidal gold, which has stronger sensitivity for detecting pathogenic bacteria and faster detection speed. DETAILED DESCRIPTION - Preparing immunochromatographic test strip marker graphene-colloidal gold involves mixing graphene powder and pyrene formic acid by weight 1-2:0.5-1.5, adding in methanol, fully reacting 1-8 hours (h) to obtain carboxylated graphene, centrifuging the carboxylated graphene, after cleaning and drying, obtain the carboxylated graphene powder, adding 30-70mM 2-(N-morpholino)ethanesulfonic acid (MES) solution to it, after 5-20 minutes (min), dropping 1-3mM 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) and 4-6mM sulfo- N-hydroxysuccinimide (sNHS) reaction, stirring under the condition of 50-100rpm for 10-40min to obtain aminated graphene, and then centrifuging for 10-40min under the condition of 5000-15000 revolutions per minute (rpm). The precipitate is resuspended, and is prepared into a 0.1-0.3mg/ml aminated graphene solution. The aminated graphene solution is placed in Sodium chloride (NaCl) solution for dialysis to desalt, dialysis for 16-28h, pure water is added, and the pH value is adjusted to 8.0-9.5. The colloidal gold solution of 0.02-0.10mg/ml is prepared and regulates its pH value to be 8.0-9.5. The aminated graphene is mixed with the colloidal gold solution for 20-60min to obtain the stone blackene-colloidal gold marker.
