▎ 摘 要
NOVELTY - A biodegradable nanocomposite comprises graphene oxide and lipid in liposome at ratio of 0.005-0.03. The graphene oxide is coated as film on inner side and outer side of liposome. The nanocomposite is functionalized. USE - Biodegradable nanocomposite. ADVANTAGE - The product is collapses on therapy and biodegradable, highly biocompatible (cell viability of more than 95%) and easily synthesized at ambient conditions even at large scale, has large hydrophilic cavity for high cargo, emits near infrared (NIR) fluorescence for easy diagnosis via deep tissue penetration, absorbs NIR (600-900 nm) for localized photothermal therapy, uses red fluorescent graphene oxide flakes (GOP), exhibits hyperthermia (crossing 43 degrees C) within 2 mm exposure of 808 nm NIR laser source with low power density (1 W/cm2), and efficiently targets cancer cell and performs targeted combined chemo-photothermal therapy. Breast cancer cells MDA-MB-231 are treated with folic acid functionalized nanocomposite. The cancer cells are seeded in 96 well plates at density of 2asterisk104 cells/well, incubating in incubator for overnight, maintaining at 5% carbon dioxide and 37 degrees C, treating cells with 100 mu l of (200 pg/mL) GOP-Lipo, GOP-Lipo-PA and DOX-GOP-Lipo-FA with and without NIR exposure for 5 mm, washing incubation wells after 24 hours with PBS, adding 20 mu l MTT dye, forming formazan crystals after 4 hours, dissolving by 200 mu L DMSO, and recording optical absorbance using microplate reader (Tecan Infinite 200 PRO) at 570 nm and 690 nm. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: (1) composition comprising nanocomposite and cargo comprising drug, biomolecule and dye; and (2) process for synthesis of nanocomposite comprising dissolving amphiphilic compound or lipid in volatile solvent to obtain mixture, removing volatile solvent from mixture, adding 0.5-3 mg phosphate-buffered saline containing fluorescent graphene oxide nanoflakes to obtain lipid suspension in form of multilamellar vesicles during film hydration process, and extruding using extruder with polycarbonate membranes at 40-50 degrees C.
