NOVELTY - CRISPR-SPR biosensor chip comprises SPR biosensor chip, Cas protein immobilized on the surface of the SPR biosensor chip, and sgRNA bound to the Cas protein; the sequence of the nucleic acid comprises a target nucleic acid sequence; the sgRNA comprises a guide sequence and an anchor sequence, the guide sequence being identical to the target nucleic acid sequence or reverse complementary to the target nucleic acid sequence, the anchor sequence specifically binds to the Cas protein. USE - The CRISPR-SPR biosensor chip, the CRISPR-SPR detection system is useful in detection of nucleic acid, and preparing of products for detecting nucleic acid (claimed). ADVANTAGE - The CRISPR-SPR biosensor chip can achieve label-free, rapid, efficient and specific detection of nucleic acids. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are also included for: (1) Preparing CRISPR-SPR biosensor chip, comprising immobilizing the Cas protein on the surface of the SPR biosensor chip by covalent bond, biotin-streptavidin, or His-tag-His antibody or His-tag-nickel-NTA, and then incubating with the sgRNA; (2) CRISPR-SPR detection system, comprising CRISPR-SPR biosensor chip, and SPR detector; and (3) Detecting nucleic acid, comprising the step of using the CRISPR-SPR biosensor chip or the CRISPR-SPR detection system.
