▎ 摘 要
NOVELTY - A composite cell membrane biomimetic targeting antibacterial nano drug delivery system comprises a liposome drug-loaded core including phospholipid, cholesterol, antibiotic and phototherapeutic agent and a composite membrane formed by red blood cell membranes and macrophage membranes coated outside the liposome drug-loaded core, where the antibiotic comprises amikacin, ciprofloxacin, levofloxacin, moxifloxacin, imipenem, aztreonam or tobramycin. USE - As composite cell membrane biomimetic targeting antibacterial nano drug delivery system. ADVANTAGE - The composite cell membrane expands and enhances the function of drug-loaded nanosystem to obtain an ideal long circulation time, reduces the degradation rate in vivo, reduces the dosage of antibiotics while improving treatment efficiency, reduces the possibility of bacterial drug resistance and systemic toxicity, prolongs the life of the drug-carrying system, realizes the slow release of antibiotics and has biomimetic properties, good biocompatibility and non-immunogenicity properties. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for a preparation method of the composite cell membrane biomimetic targeting antibacterial nano drug delivery system comprising (S1) dissolving phospholipid and cholesterol in an organic solvent, evaporating and removing the organic solvent to form a thin film, adding a mixed aqueous solution of phototherapeutic agent and antibiotic, performing hydration reaction, ultrasonically dispersing and filtering through an aqueous filter membrane to obtain liposome drug-loaded inner core loaded with antibiotics and phototherapeutic agents, (S2) separating red blood cells by low-speed centrifugation, cracking in an ice bath, performing high-speed centrifugation and freeze-drying to obtain red blood cell membranes, (S3) resuspending macrophages in tris(hydroxymethyl)aminomethan-magnesium salt buffer, leaving, sonicating in an ice bath, centrifuging, precipitating and freeze-drying to obtain macrophage membrane and (S4) dissolving the red blood cell membrane and macrophage membrane in phosphate buffered saline solution and ultrasonically dispersing to prepare a composite membrane, mixing with the liposome drug-loaded inner core, co-extruding and centrifuging to obtain the composite cell membrane biomimetic targeting antibacterial nano drug delivery system.
