DETAILED DESCRIPTION - Kit comprising microfluidic chip, purified recombinant proteins cancer antigen (CA)153, CA125, carcinoembryonic antigen (CEA), alpha-fetoprotein (AFP) and CA199, biotin-labeled CA153 monoclonal detection antibody, CA125 monoclonal detection antibody, CEA monoclonal detection antibody, AFP monoclonal detection antibody and CA199 monoclonal detection antibody, APC-streptavidin conjugate, confining solution, working solution and antibody diluent, where the microfluidic chip comprising reaction layer substrate and micro-flow channel sample adding layer on it, the reaction layer substrate comprising glass slide substrate and graphene oxide quantum dot nano material substrate assembled on glass slide substrate, and the oxidized graphene quantum dot nano material substrate is printed with CA153 monoclonal capture antibody barcode, CA125 monoclonal capture antibody barcode, CEA monoclonal capture antibody barcode, AFP monoclonal capture antibody barcode, and CA199 monoclonal capture antibody barcode, is new. An INDEPENDENT CLAIM is also included for using a kit for detecting breast cancer tumor markers, comprising (i) attaching the micro-channel loading layer is printed with CA153 monoclonal capture antibody barcode, CA125 monoclonal capture antibody barcode, CEA monoclonal capture antibody barcode, AFP monoclonal capture antibody barcode and CA199 monoclonal capture antibody barcode to the reaction layer substrate by their own weight to form a closed reaction chamber, (ii) respectively injecting different samples to be testing from multiple inflow holes, incubating for 10-40 minutes, completely combining the tumor markers in the sample to be testing with the monoclonal capture antibody on the reaction layer substrate, completing the incubating, and pumping the excess sample to be testing from the outflow hole, (iii) uncovering the film in DPBS containing 0.5-5% bovine serum albumin (BSA), and using 0.5-5% BSA to clean the tumor markers that are not adsorbed on the reaction layer substrate, (iv) taking the mixed detection antibody solution, spreading it on the reaction layer substrate, attaching a sealing film to the reaction layer substrate and incubating for 10-40 minutes, and (v) taking off the sealing film, 0.55% rinsing with BSA, taking the APC-streptavidin conjugate and spreading on the reaction layer substrate, specifically binding the APC-streptavidin conjugate to the biotin-labeled tumor marker detection antibody, fixing on the reaction layer substrate, and performing fluorescent detection of tumor markers.
