NOVELTY - Detecting cholesterol by enzyme and graphene coordinative catalyzing silver deposition method comprises e.g. (i) placing the screen printing electrode in a sulfuric acid solution and rinsing with pure water; (ii) placing activated screen printing electrode in chloroauric acid and carrying out potentiostatic deposition; and (iii) taking cholesterol and silver nitrate solution on the immobilized enzyme and graphene electrode surface, reacting drawing standard curve of cholesterol and calculating the sensitivity and detection limit. USE - The method is useful for detecting cholesterol by enzyme and graphene coordinative catalyzing silver deposition method. ADVANTAGE - The method is simple to operate and economical, saves time and has low detecting line. DETAILED DESCRIPTION - Detecting cholesterol by enzyme and graphene coordinative catalyzing silver deposition method comprises (i) placing the screen printing electrode in a sulfuric acid solution to carry out cyclic voltammetry scanning to activate the surface of the electrode to obtain the activated screen printing electrode and rinsing with pure water; (ii) placing activated screen printing electrode in chloroauric acid, carrying out potentiostatic deposition by stirring by magnetic stirrer, finishing deposition, rinsing electrode with water, placing mixture in graphite and distilled water, dispersing ultrasonically to obtain a graphene suspension, adding 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide and N-Hydroxysuccinimide solution into the suspension, adding CHER solution and CHOD solution and placing the mixed solution in a refrigerator for storage in cold storage; and (iii) taking cholesterol and silver nitrate solution on the immobilized enzyme and graphene electrode surface, placing the electrode in a 37 degrees C and dark environment, reacting for 30 minutes to obtain working electrode deposited with simple silver, washing the electrode surface with glycine- sodium hydroxide buffer, placing electrode in 0.6 mol/liter potassium nitrate- 0.1 mol/liter nitric acid solution, scanning linearly at 0.1-0.6 v at range of 100 mV/second stripping peak of silver current value according to the stripping voltammetry current value of the size of silver, drawing the standard curve of cholesterol and calculating the sensitivity and detection limit.
