▎ 摘  要

NOVELTY - BP- polyetherimide (PEI)-RNA nano medicine complex, comprises nano material with positive charge and nucleic acid with negative charge; the nucleic acid is RNA; the RNA comprises lncRNA, and circRNA. The nano-material with positive charge is nano material modified by PEI; the nano-material comprises nano-black phosphorus layer, liposome, graphene oxide, nano-gold, gelatin, silk fibroin, collagen, chitosan, and nano-polyester material; the nano material is a nano black phosphorus layer. USE - BP- polyetherimide (PEI)-RNA nano medicine complex or BP-PEI-RNA nano-drug complex with fluorescent substance label is used in treatment of disease, e.g. tumor, preferably prostate cancer (claimed). ADVANTAGE - The nano-drug complex can realize efficient and stable delivery of RNA. The exogenous gene drug (RNA) is transported to the tumor cell; by improving the expression level of cancer suppressor gene in tumor cell, exerting the function of inhibiting prostate cancer progression, it has good clinical application prospect. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are also included for: BP-PEI-RNA nano medicine complex with fluorescent substance label, comprising BP-PEI-RNA nano medicine complex and a fluorescent substance connected to the RNA; preparation method of the BP-PEI RNA nanodrug complex comprising: (1) preparing RNA; (2) preparing nano material with negative charge; and (3) incubating the RNA and the nanomaterial with negative charge; and preparing BP-PEI-RNA nanodrug complex with fluorescent substance label comprising: (a) preparing RNA with fluorescent substance label; (b) preparing nano material with negative charge; and (c) co-incubating the RNA and the nanomaterial with negative charge; preferably, the specific preparation process of step (a) is as follows: using linearized DNA of RNA as template, taking ribonucleotide labeled by fluorescent substance as fluorescent substrate, transcribing in vitro to obtain RNA marked by fluorescent substance, the specific preparation process of step (b) is as follows: taking the nano black phosphorus layer solution, centrifuging and removing the supernatant, re-suspending using enzyme-free water, mixing uniformly, ultrasonicating, repeatedly centrifuging, enzyme-free water re-suspending, ultrasonicating to obtain the nano black phosphorus layer aqueous phase dispersing solution; then adding PEI solution to the aqueous phase dispersion solution of the nano-black phosphorus layer for ultrasound; removing the excess polyetherimide by centrifugal washing and re-suspending it; the nano black phosphorus layer aqueous phase dispersion solution with PEI solution volume ratio is 1: 1.5; the ultrasonic is ice water bath ultrasonic; the specific preparation process of step (b) is as follows: taking 1 ml nano black phosphorus layer solution with concentration of 0.1 mg/ml in the EP tube, centrifuging for 20 minutes at 4℃ at 16000 revolutions/minute (rpm), removing the supernatant, equal amount of enzyme free water re-suspending, pulping and mixing uniformly at 40 kHz, 100 W ice water bath ultrasonication for 20 minutes, repeating centrifuging, enzyme-free water re-suspending, ice water bath ultrasonication step for 2 times to obtain the dispersed nano black phosphorus layer aqueous phase dispersing solution; then adding 2.5k in the nano black phosphorus layer aqueous solution, the concentration is 10 mg/ml PEI, nano black phosphorus layer and PEI volume ratio is 1: 1.5, ice water bath ultrasonication for 3-4 hours; centrifugally washing to remove the excess PEI and re-suspending it; the specific preparation process of step (c) is as follows: obtaining the nano black phosphorus layer solution modified by PEI through step (b), adding RNA with fluorescent substance label, co-incubating, continuously rotating and mixing uniformly to obtain the BP-PEI-RNA nano-drug complex with fluorescent substance label; the mass ratio of the nano black phosphorus layer and RNA with fluorescent substance label is 1: 0.4; the specific preparation process of step (c) is as follows: adding RNA with fluorescent substance label, so that the mass ratio of the nano black phosphorus layer and the fluorescent substance label is 1: 0.4, incubating for 10 minutes at 4 ℃, continuously rotating and mixing uniformly to obtain the BP-PEI-RNA nano medicine complex.
