NOVELTY - Preparing graphene-chitosan-silver nitrate complex (A) comprises (i) preparing graphene-chitosan suspension, and (ii) mixing the graphene-chitosan suspension with silver nitrate aqueous solution, and ultrasonic reacting and heating to obtain the complex A. The preparation of the graphene-chitosan suspension comprises (a) dissolving chitosan in aqueous glacial acetic acid solution and mixing to obtain the chitosan solution, and (b) mixing the chitosan solution with graphene and ultrasonic processing. USE - The complex is useful for preparing reovirus detecting electrochemical immunosensor (claimed). ADVANTAGE - The electrochemical immunosensor prepared by the complex, does not require the addition of electrochemical active substance as an electrochemical indicator into a testing base solution. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are also included for: (1) complex A prepared by the above method; (2) preparing the complex B, comprising reacting complex A with bovine serum albumin aqueous solution of antibody to obtain complex B; (3) complex B prepared by the above method; (4) a kit useful for preparing electrochemical immunosensor working electrode, comprising independently packed complex B and complex C, where preparation of the complex C comprises mixing chloroauric acid aqueous solution with graphene-chitosan suspension, heating and reacting to obtain the complex C, the ratio of chitosan, glacial acetic acid, graphene and chloroauric acid is 5 g:28 ml:1 g:0.2 g, the mixing temperature is 25-28 degrees C, preferably 28 degrees C and the time is 30-120 minutes, preferably 30 minutes, the reaction temperature is 80 degrees C and the time is 0.5-2 hours, preferably 1 hour; and (5) preparing the working electrode useful for detecting reovirus electrochemical immunosensor, comprising (a) coating complex C of the kit on surface of the gold electrode and drying to obtain complex C modified gold electrode, (b) coating the sample to be tested on the complex C modified gold electrode and reacting to obtain the detection test sample fixed gold electrode, (c) coating the aqueous solution of bovine serum albumin on the detection test sample fixed gold electrode and reacting to obtain bovine serum albumin sealed gold electrode, and (d) coating the complex B on the bovine serum albumin sealed gold electrode and reacting to obtain the working electrode, where in the step (b), the reaction temperature is 2-8 degrees C, preferably 4 degrees C and the reaction times is 8-24 hours, preferably 8 hours, in the step (c), the reaction temperature is 2-8 degrees C, preferably 4 degrees C and the reaction times is 8-24 hours, preferably 12 hours, and in the step (d), the reaction temperature is 37 degrees C, and the reaction times is 30-60 minutes, preferably 40 minutes; and (6) sleeved product useful for electrochemical immunosensor detection of the sample to be tested, comprising complex B and immunosensor, where the immune sensor is gold electrode, preferably complex C modified gold electrode of the kit, or electrochemical immunosensor working electrode, or the working electrode containing the complex A, complex B and the kit which is suitable for detecting the reovirus.
