NOVELTY - A multi-functional graphene sensor preparing method involves polishing glassy carbon electrode with alumina powder. The polished electrode is added with mercapto based graphene aqueous solution and dried. The modified electrode surface is added with capture antibody. The cleaned electrode is added with bovine serum albumin in non-specific active sites, incubated and cleaned, followed by adding melanoma antigen, adding multi-functionalized graphene labeled melanoma tumor detection antibody solution on the electrode and antigen-specific recognition, incubating and storing in refrigerator. USE - Method for preparing multi-functional graphene sensor for detecting melanin tumor (claimed). ADVANTAGE - The sensor has high sensitivity. DETAILED DESCRIPTION - A multi-functional graphene sensor preparing method involves polishing glassy carbon electrode with 1.0, 0.3 and 0.05 mu m alumina powder, cleaning ultrasonically with water and ethanol and drying using nitrogen. The polished electrode is added with 1-3 mg/ml mercapto based graphene aqueous solution and dried to obtain modified electrode surface. The modified electrode surface is added with 8-12 mu g/ml capture antibody solution dropwise, incubated in refrigerator at 4 degrees C for 1 hour and cleaned. The cleaned electrode is added with 5-5 mg/ml bovine serum albumin solution in non-specific active sites, incubated in refrigerator at 4 degrees C for 1 hour and cleaned, followed by adding 6 mu l melanoma antigen of series of different concentration of 0.00001-100 mg/ml for capturing antibody of specific recognition, adding 6 mu l multi-functionalized graphene labeled melanoma tumor detection antibody solution on the electrode and antigen-specific recognition, incubating at room temperature for 1 hour and storing in refrigerator at 4 degrees C. An INDEPENDENT CLAIM is also included for a method for detecting melanin tumor, which involves selecting three-electrode electrochemical workstation system for testing, taking saturated calomel electrode as reference electrode, molybdenum wire electrodes as auxiliary electrode, multi-functional graphene sensor as working electrode, adding 10 ml phosphate buffer solution, selecting chronoamperometry melanoma antigen detection with input voltage of -0.4 V, sampling at interval of 0.1 second and running time of 400 seconds, injecting sample in phosphate buffer solution after 50 seconds of background current stabilized, recording current changes with time, drawing working curve and testing melanin tumor.
