NOVELTY - Method for preparing electrochemical gene sensing device, involves (a) preparing platinum-gold-based three-dimensional graphene (Pt-Au-3DGR) nano-compound material, (b) preparing carbon ionic liquid electrodes (CILE), (c) preparing modified electrode, and (d) preparing electrochemical gene sensing device by immobilizing single-stranded DNA (ssDNA) probe on surface of modified electrode, hybridizing, washing the electrode, and designating the electrode as double stranded DNA (dsDNA)/Pt-Au-3DGR/CILE. USE - The method is useful for preparing electrochemical gene sensing device for detecting gene sequence (claimed). ADVANTAGE - The constructed electrochemical gene sensing device exhibits good selectivity and high sensitivity. DETAILED DESCRIPTION - Method for preparing electrochemical gene sensing device, involves (a) preparing platinum-gold-based three-dimensional graphene (Pt-Au-3DGR) nano-compound material by mixing 2.5 ml chloroauric acid and 2.426x 10-2 mol/l chloroplatinic acid in a ratio of 1:1 for 15 minutes, adding suitable amount of graphene (GO) solution, and distilled water, stirring uniformly for 15 minutes, transferring the mixture to 50 ml polytetrafluoroethylene reactor, heating mixture at 180 degrees C for 12 hours, cooling mixture to room temperature, removing the precipitate, washing twice with distilled water for 24 hours, and freeze-drying mixture at -80 degrees C for 12 hours to obtain Pt-Au-3DGR nano-compound material, (b) preparing carbon ionic liquid electrodes (CILE) by mixing graphite powder, ionic liquid and liquid paraffin, adding mixture into glass tube, and inserting copper wire with iron bars for compactness to obtain CILE, and polishing CILE, (c) preparing modified electrode by modifying surface of CILE and nano-compound material by dropping method at room temperature, and (d) preparing electrochemical gene sensing device by immobilizing single-stranded DNA (ssDNA) probe on surface of modified electrode by potentiostatic adsorption, adding 10 mu l solution containing target ssDNA sequence on the surface of the electrode by dropping method, subjecting the target ssDNA sequence to molecular hybridization with the probe ssDNA sequence at room temperature, washing the electrode with 5% sodium dodecyl sulfate (SDS) solution and secondary distilled water after 30 minutes of hybridization, and designating the electrode as double stranded DNA (dsDNA)/Pt-Au-3DGR/CILE.
