NOVELTY - Method for screening Rev polypeptide small molecule antagonist based on graphene oxide, involves (a) adding the pro-flavin and HIV-I rev response element (RRE) RNA fragment having IIB stem loop portion to a buffer and incubating at 37 degrees C for 1-5 minutes, (b) adding the test drug and incubating at 37 degrees C for 1-5 minutes, (c) adding graphene oxide and uniformly mixing mixture, and (d) measuring fluorescence polarization degree of the reaction system before and after addition of the test drug. USE - The method is useful for screening Rev polypeptide small molecule antagonist. ADVANTAGE - The method eliminates possible defects in conformational changes of macromolecules, performs high-throughput screening of Rev polypeptide small molecule antagonist with high sensitivity. DETAILED DESCRIPTION - Method for screening Rev polypeptide small molecule antagonist based on graphene oxide, involves (a) adding the pro-flavin and HIV-I rev response element (RRE) RNA fragment having IIB stem loop portion to a buffer having pH of 6.5-8 and incubating at 37 degrees C for 1-5 minutes, (b) adding the test drug and incubating at 37 degrees C for 1-5 minutes, (c) adding graphene oxide and uniformly mixing mixture, and (d) measuring fluorescence polarization degree of the reaction system before and after addition of the test drug, if the test drug has an antagonistic effect then the degree of polarization is significantly reduced and the Rev polypeptide is modified in accordance for screening of molecular antagonists.
