NOVELTY - Cartridge comprises first compartment configured for performing cell transfection, where first compartment comprises a first inlet configured for introduction of a cell sample; a second compartment configured for performing cell selection, where an inlet of the second compartment is operably coupled to an outlet of the first compartment, and the second compartment further comprises one optically-transparent wall and an outlet that is operably coupled to an intermediate cell removal port; and a third compartment configured for performing cell expansion, where an inlet of the third compartment is operably coupled to the outlet of the second compartment. USE - Cartridge used in system, and apparatus for producing clonal population of transfected cells. ADVANTAGE - The cartridge ensure significant reduction in the total number of cells required as input for generation of clonal cell populations, reduction in the total number of cells required to be processed for generation of clonal cell populations, in cell culture reagent and consumables consumption, improvements in cell transfection efficiency, improvements in the overall efficiency of generating clonal cell populations, and higher overall throughput (i.e., reduced start to finish times) for generating clonal cell populations. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for the following: (1) system comprises a cartridge comprising one compartment configured for performing cell transfection, cell selection and/or cell expansion, where the cartridge comprises an inlet configured for introduction of a cell sample and the one compartment comprises an optically-transparent wall; (2) method for producing a clonal population of transfected cells, involves providing a cartridge, where the cartridge comprises one compartment configured for performing cell transfection, cell selection, cell expansion, or any combination, and compartment comprises an optically-transparent wall; introducing a cell sample into the one compartment; transfecting the cell sample with one or more transfection agents; selecting one clonal cell colony derived from the transfected cell sample; performing photoablation to destroy all clonal cell colonies except the one clonal cell colony selected in; and subjecting the one clonal cell colony selected to one or more cycles of cell division and growth to produce a clonal population of transfected cells; (3) apparatus comprises cartridge, where cartridge comprises one compartment configured for performing cell transfection, cell selection, cell expansion, or any combination, and compartment comprises an optically-transparent wall that is operably coupled to a source of laser light for performing photoablation and photodetachment, and a controller; and (4) non-transitory computer-readable medium storing a set of instructions which, when executed by a processor, cause a processor-controlled system to perform controlling timing and flowrate for one or more fluids flowing through a cartridge comprising compartment configured to perform cell transfection, cell selection, cell expansion, or any combination; performing image processing of images acquired by an imaging unit configured to image a surface or volume within the compartment and, based on data derived from the processed images, selecting a first subset of cells growing on a surface of or in a volume within the compartment for laser-based photodetachment and second subset of cells growing on a surface of or in a volume within the compartment for laser-based photoablation; and controlling one or more operating parameters of one or more lasers and a laser targeting unit such that the first subset of cells is photodetached and the second subset of cells is photoablated.
