NOVELTY - Graphene culture substrate comprises a supporting layer, a graphene layer and a poly methyl methacrylate (PMMA) layer from bottom to top. USE - The graphene culture substrate is useful for culturing, differentiating and transplanting pluripotent stem cell and inducing pluripotent stem cell regeneration of cardiac cell in the implantation and transplantation therapy (claimed) of heart failure patient and arrhythmia. ADVANTAGE - The graphene culture substrate can be adjusted in size according to actual needs. The cultured mature myocardium is suitable for drug detection and development, disease model research and clinical heart disease treatment. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for the following: (1) method of preparing the culture substrate involves (a) introducing hydrogen in a quartz tube, performing heat treatment of a copper substrate, then passing methane and preparing a graphene film by chemical vapor deposition method on a copper substrate, turning off the methane at the end of the growth, naturally cooling to room temperature, turning off the hydrogen gas, releasing the gas until the pressure inside and outside the quartz tube is equal and removing the graphene/copper, (b) spin-coating 2-10% PMMA on the graphene/copper surface using a spin coater and placing the sample in an ammonium persulfate solution to remove copper by etching, and (c) transferring the PMMA/graphene to a clean supporting layer, slowly blowing with a low-flow nitrogen gas gun to remove the surface moisture and then air-drying for 0.5-10 hours to completely remove the water to obtain the product; and (2) use of the graphene culture substrate for culturing and differentiating the induced pluripotent stem cell and inducing pluripotent stem cell regeneration of cardiac cell in the implantation and transplantation therapy.
