NOVELTY - Microtip device has an array of microtips, where each microtip comprises a first layer of reduced Graphene Oxide (rGO) mixed Polydimethylsiloxane (PDMS) forming the tip and a second layer of PDMS forming the base. USE - Microtip device for use in cellular diagnostics, biomedical research, and disease intervention (claimed). ADVANTAGE - The device utilizes for efficient intracellular delivery of biomolecules with high transfection efficiency and cell viability, employs direct intracellular delivery, and has ability to absorb surrounding proteins and good biocompatibility. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: (1) a method for preparing a rGO mixed PDMS microtip device, which involves: (a) fabricating a microtip device mold; (b) preparing a rGO mixed PDMS mixture by mixing photo-reduced graphene oxide (rGO), PDMS and a curing agent in a weight ratio of 1:10:1; (c) pouring said rGO mixed PDMS mixture onto the microtip device mold followed by PDMS and curing at a temperature of 50-60 °C overnight; and (d) peeling off the rGO mixed PDMS microtip device from the Si mold after 48 hours to obtain the rGO mixed PDMS Microtip device; and (2) a method for intracellular delivery of molecules by rGO mixed PDMS microtip device, which involves: (a) aligning the rGO mixed PDMS microtip device placed on top of monolayer of cells in such a way that microtip device is in contact with the cells; (b) exposing nanosecond pulse laser with 10 Hz frequency on the aligned device for creating photothermal bubbles and formation of membrane pores; and (c) delivery of the biomolecules through said membrane pores into the cell.
