NOVELTY - Shewanella with graphene affinity, is new. The Shewanella MR-1 whose mtrC gene has been knocked out is used as a host, and the plasmid DNA contains the graphene affinity peptide domain and the gene fragment mtrC-EPLQLKM expressing the MtrC protein. USE - The Shewanella with graphene affinity is useful in field of reducing graphene oxide (claimed). DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for constructing Shewanella with graphene affinity comprising (1) translating peptide domain EPLQLKM into a nucleotide sequence, and connecting flexible polypeptide GSGGSG to the C-terminus of the Shewanella MR-1 extracellular protein mtrC gene, and adding flexible polypeptide GS before the terminator to constructing expression MtrC protein gene fragment mtrCEPLQLKM, (2) adding restriction sites and protective bases to the N-terminus and C-terminus of the gene fragment mtrCEPLQLKM, and amplifying mtrC-EPLQLKM product by PCR, (3) connecting mtrC-EPLQLKM product with the expression vector pBBRParcA by double enzyme digestion to construct a recombinant plasmid pBBR-ParcA-mtrCEPLQLKM, (4) transforming constructed recombinant plasmid pBBRParcA-mtrC-EPLQLKM into host Escherichia coli WM3064, and combining and transferring to Shewanella MR-1 knocked out of the mtrC gene, constructing into recombinant Shewanella, both Shewanella with graphene affinity.
