▎ 摘  要

NOVELTY - Utilizing multifunctional graphene-carrying net involves etching copper foil covered by graphene film grown by chemical vapor deposition (CVD) as the electric mirror carrying net to obtain the graphene carrying net. The electrode carrying net is provided with a hole array. The oxygen plasma treatment is performed on the graphene carrier net. The 2-(N-morpholino)ethylanesulfonic acid buffer solution is used to process the graphene carried net, where the 2-N-methylsulfonate buffer solution contains 1-ethyl-3-(3-dimethyl-amino propyl)carbodiimide hydrochloride and N-hydroxysulfosuccinimide. The ammonia triacetic acid and biotin activation is used for the treatment of the graphene carry net. USE - Method for utilizing multifunctional graphene-carrying net used in three-dimensional reconstruction of freezing electron microscope used in structural biological analysis. ADVANTAGE - The method enables to prepare multifunctional graphene-carrying net, which can specifically anchor the target biomolecule on the surface, so as to avoid the interface problem of air and water, and can control the particle orientation distribution, through the related protein purification tag is constructed on different directions of each biological macromolecule, providing a rich Euler angle, and the structure analysis of the biological macro-molecule is more reliable and more efficient, thus, MFG carrier network promotes the commonality and repeatability of application of freezing electron microscope in structural biological analysis.
