▎ 摘 要
NOVELTY - Detecting biotin comprises (i) taking 0.5-2 g sample into centrifugal tube, adding 0.9 ml internal standard intermediate liquid and adding ultra-pure water at temperature of 14.1 ml/50, uniformly mixing, shaking, adding 60% perchloric acid solution and 5 ml dichloromethane, vibrating, centrifuging, taking out supernatant, adding ultra-pure water and perchloric acid solution, extracting for one time, and combining two times of supernatant to obtain liquid to be purified, and (ii) using pillared layered structure (PLS) ion exchange column, orderly adding methanol, water activated small column, adding purified liquid into PLS ion exchange column, using 6 ml de-ionized water to wash, removing effluent liquid, using methanol to wash, collecting washing liquid, drying by nitrogen, using water to dissolve, shaking and uniformly mixing, using aqueous phase filter membrane for filtering, using liquid chromatography-tandem mass spectrometry (LC-MS/MS) instrument for analysis and detection. USE - The method is useful for detecting biotin. ADVANTAGE - The method: improves the aqueous phase filter film in the filtering process; achieves the bacteriostatic effect; prepares the reinforcing filler in the aqueous phase filter film preparation process; uses nano titanium dioxide and graphene oxide composite material as carrier; enhances the toughness of the aqueous phase filter membrane; has quaternary ammonium salt structure; reduces the activity of the metabolic enzyme in the microorganism cell to block the respiration; at the same time, interferes with the production of the substance needed by bacteria production; destroys the synthesis of the cell wall, so as to kill the cell; does not reduces the antibacterial property of the aqueous phase filter film after multiple use; and ensures the accuracy of the biotin detection.
