NOVELTY - Preparing graphene chip comprises (i) preparing a gold nanoparticle solution, (ii) taking an equal amount of horseradish peroxidase HRP solution, gold nanoparticles AuNPs solution and the second antibody Ab2 solution, and mixing and incubating for 1-3 hours, adding bovine serum albumin BSA solution and incubating again to obtain the AuNPsHRP-Ab2 solution for preparing the main nanoprobe, and storing at 3-5℃, and (iii) pickling the graphene chip electrodes, sequentially immersing in a mixed solution containing 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide and N-hydroxysuccinimide, washing first antibody Ab1 solution, human epithelial growth factor receptor-2 (HER2) solution and bovine serum albumin solution after incubation to prepare the graphene chip electrode modified by Ab1-HER2, immersing graphene chip electrode modified by Ab1-HER2 in the AuNPs-HRP-Ab2 solution, and washing after incubation to prepare graphene chip electrode modified by Ab1-HER2-Ab2-HRP-AuNPs.
