NOVELTY - Preparing immobilized biological enzyme involves dispersing the graphene oxide in PBS and performing ultrasonic dispersion for more than 4h to form a uniformly dispersed suspension to obtain a graphene oxide dispersion, the concentration of the graphene oxide dispersion is 0.1-1mg/mL. The graphene oxide dispersion is placed at room temperature and 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride and N-hydroxysuccinimide are added. The amount of 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride added is 3-5 times the mass of graphene oxide. The added amount of N-hydroxysuccinimide is 3-5 times the mass of graphene oxide, stirred for 0.5-4h, then add the amination reagent, shaken at room temperature for more than 12h to amination of the graphene oxide surface, centrifuged, wash the precipitate, adding PBS buffer solution to the resulting precipitate and dispersed by ultrasound to prepare a dispersion of aminated graphene oxide. USE - Method for preparing immobilized biological enzyme. ADVANTAGE - The method enables to prepare immobilized biological enzyme, which has uniform particle size, and has good stability to high temperature, strong acid and alkali. DETAILED DESCRIPTION - Preparing immobilized biological enzyme involves dispersing the graphene oxide in PBS and performing ultrasonic dispersion for more than 4h to form a uniformly dispersed suspension to obtain a graphene oxide dispersion, the concentration of the graphene oxide dispersion is 0.1-1mg/mL. The graphene oxide dispersion is placed at room temperature and 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride and N-hydroxysuccinimide are added. The amount of 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride added is 3-5 times the mass of graphene oxide. The added amount of N-hydroxysuccinimide is 3-5 times the mass of graphene oxide, stirred for 0.5-4h, then add the amination reagent, shaken at room temperature for more than 12h to amination of the graphene oxide surface, centrifuged, wash the precipitate, adding PBS buffer solution to the resulting precipitate and dispersed by ultrasound to prepare a dispersion of aminated graphene oxide. The aminated graphene oxide dispersion is centrifuged, washed to obtain an aminated graphene oxide, and the aminated graphene oxide is dissolved in PBS. Glutaraldehyde is added after ultrasonic dispersion, and the volume-to-mass ratio of glutaraldehyde to graphene is 0-0.1ml/mg. After stirring at room temperature for 0.5-4h, biological enzyme aqueous solution is added to it, the quality of the biological enzyme is 0.1-5 times of the aminated graphene oxide, stirred at room temperature for 0.1-6h to covalently fix the biological enzyme on the surface of graphene oxide, washed and dried to obtain the immobilized biological enzyme.
