NOVELTY - Electrochemical paper-based sensor comprises a reduced graphene oxide-gold nanoparticles and poly (3,4-ethylene dioxythiophene) poly (styrene sulfonate) (PEDOT: PSS) fabricated on a cellulose fiber paper (CFP). USE - The electrochemical paper-based sensor for is useful for electrochemical sensing of bacterial infection specific biomarker (claimed). ADVANTAGE - The sensor: has a linear range sensitivity PCT of 1x 103-6x 107μ g/ml; utilizes PEDOT: PSS which has electrochemical activity, low absorption coefficients, high electrical conductivity, good ecological consistency, and visibility; and utilizes gold nanoparticles which has high sensitivity, biocompatibility, excellent stability, and superior conductivity. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for preparing the paper-based sensor comprising (A) preparing reduced graphene oxide-gold nanoparticles-PEDOT: PSS nanocomposite (i) mixing graphite powder with 1 g silver nitrate in 45 ml sulfuric acid, stirring the mixture for 3 hours of at 50° C, resuspending the same solution and then at 98° C with 100 ml of a solution containing 6 g potassium permanganate, (ii) after adding hydrogen peroxide, the solution's color turns from brown to yellow, (iii) adding 2.28g of sodium borate as a reducing agent, adding 0.5 g calcium chloride as a catalyst, and adding 0.3 mg/ml graphene oxide, stirring the solution for 8-10 hours, (iv) storing at 4° C for further use. In the second step preparing PEDOT: PSS ethylene glycol solution comprising (v) preparing an aqueous suspension of 2.4 wt.% PEDOT: PSS and 5% ethylene glycol and stirring for 30 minutes, (vi) storing reduced graphene oxide-gold nanoparticles-PEDOT: PSS nanocomposite at 4° C for further use. (B) Preparing biosensor comprising cleaning CFP electrode surface with the surfactant Tween 80(RTM: Polysorbate 80) and ethanol, repeating the procedure three times before the CFP electrode is dried, coating the dried CFP electrode with a previously prepared 5 μ l reduced graphene oxide-gold nanoparticles-PEDOT: PSS nanocomposite, and dropping 10 μ l 0.07 mg/ml anti-PCT onto the previously mentioned coated ECP electrode to immobilize the antibody, which is then dried, dropping 10 μ l of 0.25 % blocking agent bovine serum albumin and drying in air for 30 minutes for non-specific binding.
