NOVELTY - Preparing diglyceride comprises e.g. (i) adding free lipase solution into a pH 8 phosphate buffer solution whose mass fraction is 5-50%, collecting the supernatant enzyme solution, adding 30-200% graphene oxide, controlling temperature at 45 degrees C, constant temperature vibration absorbing for 6-12 hours, filtering, using phosphoric acid buffer solution to wash until there is no protein washed out to obtain graphene immobilized lipase, (ii) taking oil and 1-20% graphene immobilized lipase in any biochemical device suitable for enzyme reaction, and uniformly mixing. USE - The method is useful for preparing diglyceride (claimed). ADVANTAGE - The method has high reaction activity, mild reaction condition, can easily separate catalyst and realizes recycling. Oil prepared by diglyceride reduces visceral fat, inhibits weight gain, and reduces blood fat. DETAILED DESCRIPTION - Preparing diglyceride comprises (i) adding free lipase solution into a pH 8 phosphate buffer solution whose mass fraction is 5-50%, collecting the supernatant enzyme solution, adding 30-200% graphene oxide, controlling temperature at 45 degrees C, constant temperature vibration absorbing for 6-12 hours, filtering, using phosphoric acid buffer solution to wash until there is no protein washed out to obtain graphene immobilized lipase, (ii) taking oil and 1-20% graphene immobilized lipase in any biochemical device suitable for enzyme reaction, uniformly mixing, placing in a constant temperature oscillator, where speed is 500 revolutions/minute, controlling temperature at 30-65 degrees C, reacting for 1-10 hours to obtain the reaction product, and (iii) recycling the immobilized lipase, distilling reaction product and purifying to obtain the diglyceride.
