NOVELTY - Preparation of polyethylene glycol-modified nano-graphene oxide drug delivery system involves dissolving methoxy polyethylene glycol, carbodiimide hydrochloride and N-hydroxysuccinimide in N,N-dimethylformamide, ultrasonically processing, stirring, reacting for 0.5-1.5 hours, obtaining a reaction solution, adding doxorubicin and triethylamine, stirring at room temperature, placing the reaction solution in a dialysis bag, dialyzing with distilled water, freeze-drying, obtaining polyethylene glycol-amide-doxorubicin (A), preparing nano-graphene oxide (B) by Hummer's method using multiple layers of graphene oxide, concentrated sulfuric acid, potassium permanganate, hydrogen peroxide solution and sodium hydroxide, mixing an aqueous solution of component (A) and an aqueous solution of component (B) in mass ratio of 3:1, ultrasonically processing, stirring for 12-16 hours, centrifuging, washing, removing free doxorubicin hydrochloride, and redissolving lower layer precipitate. USE - Preparation of polyethylene glycol-modified nano-graphene oxide drug delivery system. ADVANTAGE - The method enables simple preparation of polyethylene glycol-modified nano-graphene oxide drug delivery system under mild reaction condition, without performing chemical modification of nano-graphene oxide with high drug loading amount in drug carrier of more than 100%. DETAILED DESCRIPTION - The mass ratio of graphene oxide and potassium permanganate is 1:2.5-3.5. Component (B) is obtained by adding 0.4-0.5 g multiple layers of graphene oxide to a reactor, adding 10 ml concentrated sulfuric acid, stirring, placing resultant in ice water bath, stirring, adding potassium permanganate, stirring at 0.5-1 hour, heating at 32-38 degrees C for 1.5-2.5 hours, adding deionized water, placing resultant in an oil bath, processing at 90-105 degrees C for 1-2 hours, ultrasonically processing, adding hydrogen peroxide solution, stirring, reacting for 20-40 minutes, adjusting pH using sodium hydroxide to 6.5-7, dialyzing using pure water and freeze-drying. The centrifuging is carried out at 8000-13000 rpm for 5-15 minutes.
