NOVELTY - Fluorescence based diagnostic kit comprises amine functionalized nitrogen doped graphene quantum dots (amine-N-GQDs) conjugated with anticancer cell-specific biomarker antibody as fluorescence donor and gold nanoparticles (AuNPs) as fluorescence quencher, where the binding of the anticancer cell specific biomarker antibody conjugated amine-N-GQDs to the cancer cell biomarker recovers back the fluorescence quenched by the gold nanoparticles. USE - The kit is useful for detecting presence of cancer cells and human neuron specific enolase levels in whole blood and serum from patients with small cell lung carcinoma (all claimed). ADVANTAGE - The kit: detects 0.1 pg/ml to 1000 ng/ml human neuron specific enolase with higher sensitivity at more rapid rate in maximum of 35 minutes; is economical as compared to conventional kit; is reliable and highly sensitive response with minimum background noise; is easy to operate; has capability to detect cancer in real-time; can work as an alternative to the standard enzyme linked immunosorbent assay kits for cancer biomarker detection; and utilizes GQDs which are non-toxic, environmentally friendly, chemical inertness, good water solubility, ease of modification, and good photochemical stability. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for making fluorescence based diagnostic kit, comprising (a) in situ synthesis of amine-N-GQDs by one-pot bottom-up hydrothermal synthesis, (b) synthesizing biofunctionalized graphene quantum dots (anti-NSE/amine-N-GQDs) by covalent immobilization of monoclonal neuron-specific enolase antibodies to the amine-N-GQDs synthesized in step (a), and (c) making the fluorescence based diagnostic kit utilizing anti-NSE/amine-N-GQDs as fluorescence donor and gold nanoparticles as fluorescence quencher for neuron specific enolase (NSE) detection in small cell lung cancer cells.
