NOVELTY - System (10) comprises insulator-based dielectrophoresis device (12) which further comprises fluid flow channel (14) defined by first substrate surface (22) and second substrate surface spaced from first substrate surface, where fluid flow channel have fluid inlet (16) and outlet, insulating flow structure extending from first substrate surface toward second substrate surface by defining constriction in fluid flow channel between first and second substrate surface; detection chamber placed in fluid communication with fluid flow channel by opening in first or second substrate surface, where opening is configured downstream of at least one insulating flow structure, detection chamber includes electrochemical sensor configured to constrict flow of fluid entering detection chamber through pore, where pore is sized to produce detectable signal upon passage of analyte through pore; electrodes in electrical communication with fluid channel inlet and outlet of fluid flow channel. USE - System used for separating and characterizing first analyte from second analyte in fluid mixture (claimed), separating fluid mixture contains multiple analytes using gradient insulator-based dielectrophoresis (g-iDEP) into selected individual analytes and transporting selected analyte to nanopore in device for characterization or detection. ADVANTAGE - The device enables to provide easy portability and use on wide range of analyte includes bioparticle; allow for smart, electrically-tuned single-molecule sensing in addition to simultaneous separation of complex fluid mixture; allow for simultaneous separation, concentration, characterization or detection of analyte of interest. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for a method for separating, and characterizing first analyte from at least second analyte in fluid mixture, which involves: (a) transporting fluid mixture, which comprises first analyte, second analyte through system; (b) separating second analyte from first analyte by passing fluid mixture through constriction; and (c) transporting first analyte through pore of electrochemical sensor to produce detectable signal.
