NOVELTY - Preparing silver-bismuth hybrid based electrochemiluminescence immunosensor involves polishing the glassy carbon electrode with alumina powder, followed by rinsing the polished electrode with ultrapure water. 6microliters Aminated graphene-gold/ceria particles containing solution (1-3 mg/mL) is dripped on the surface of polished electrode, followed by rinsing the electrode with ultrapure water. 6microliters Antibody solution (5-10microg/mL) is dripped on the surface of electrode, followed by drying, and rinsing the same with ultra-pure water. USE - Method for preparing silver-bismuth hybrid based electrochemiluminescence immunosensor used for detecting prostate specific antigens (claimed). DETAILED DESCRIPTION - Preparing silver-bismuth hybrid based electrochemiluminescence immunosensor involves polishing the glassy carbon electrode with alumina powder, followed by rinsing the polished electrode with ultrapure water. 6microliters Aminated graphene-gold/ceria particles containing solution (1-3 mg/mL) is dripped on the surface of polished electrode, followed by rinsing the electrode with ultrapure water. 6microliters Antibody solution (5-10microg/mL) is dripped on the surface of electrode, followed by drying, and rinsing the same with ultra-pure water. The electrode surface is then freeze-dried in a refrigerator at 4 degrees C. The antibody is incubated with 3microliters of bovine serum albumin solution (1%) in refrigerator at 4 degrees C for 3hours for blocking the non-specific enzyme active site of the antibody. The electrode is then uniformly washed. 6microliters Prostate specific antigen is added on the electrode for performing antigen-antibody capture reaction. Antigen-antibody capture reaction is performed by incubating the reaction mixture at room temperature for an hour, followed by cleaning the electrode. 6microliters concentration of silver-bismuth sulfide hybrid solution (1-3 mg/mL) is transferred to antigen antibody labeled-electrode surface, and then incubated at room temperature for an hour, followed by cleaning, and then storing at 4 degrees C under refrigerated condition. An INDEPENDENT CLAIM is included for a method for detecting prostate specific antigens, which involves: (A) using silver-bismuth hybrid based electrochemiluminescence immunosensor; (B) performing detection process in an electrochemical luminescence system in presence of 100-150mmol/L potassium persulfate buffer solution with the prepared silver-bismuth hybrid based electrochemiluminescence immunosensor; and (C) using the electrochemical luminescence system to detect the intensity of electrochemical luminescence signal produced by different concentration of antigen, and then drawing the curve based on test results.
