NOVELTY - The modulation, and signal detection device comprises a microfluidic top layer that is provided with a first groove, a sampling flow passage, and a sampling micro-valve for controlling opening, and closing of the sample outlet flow passage. A micro-flow control bottom layer forms with a second groove. A control circuit contains a first power supply, and a signal generator electrically connects with the first power source. A first control electrode, and a second control electrode arranges on the micro-fluidic control top layer, and adapted to contact with a sample solution to be tested. A detection circuit collects a graphene nano- hole facing electric signal equal to a reference frequency. USE - Single-molecule periodic via modulation, and signal detection device based on nanopore technology set. ADVANTAGE - The sample translocation behavior in the nano- hole detection randomness is improved. The detection of easy bit event under the single molecular scale, and detection of weak signal is realized.
