NOVELTY - Graphene mixed filler comprises graphene materials and chromatography medium fillers in a weight ratio of 1-50:100. The particle size of the graphene material is 500-5000 mesh, and particle size of the chromatography medium filler is 200-500 mesh. USE - The graphene mixed filler is useful in graphene mixed packing chromatography column for separation and purification of compound medicine with pi -conjugated electronic structure including alkene and alkyne structure (all claimed). ADVANTAGE - The graphene mixed filler: has graphene ( pi - pi electron conjugation effect) to achieve column adsorption of compounds with a pi -conjugated electronic structure, adsorbs impurities, avoids multiple operations, and reduces chromatography cost and operation time. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are also included for: (1) preparing graphene mixed filler comprising adding graphene suspension into chromatographic medium filler having particle size of 200-500 mesh, and mixing uniformly, drying at 90-100 degrees C for 2-5 hours to obtain graphene mixed filler, where weight ratio between graphene in the graphene mixed filler and the chromatography medium filler is 1-50:100, and chromatographic medium filler includes bonded silica gel, alumina, diatomaceous earth or cellulose powder, and graphene suspension is by adding water having concentration of 45-55 wt.% ethanol aqueous solution into a graphene material having particle size of 500-5000 mesh, and then uniformly stirring; and (2) graphene mixed packing chromatography column comprising chromatography column divided into upper part and a lower part, and filling upper part of the chromatography column with the graphene mixed filler, and filling lower part of the chromatography column with chromatography medium filler used in upper part of the chromatography column, and filler height at the lower part of the chromatography column is 5-10% the total filler height of the chromatography column.
