▎ 摘 要
NOVELTY - Preparing hyaluronic acid/graphene oxide nanocomposite hydrogel comprises preparing tyramine grafted hyaluronic acid by adding hyaluronic acid into 2-(N-morpholino)ethanesulfonic acid buffer solution, mixing uniformly, adding ethyl(dimethylaminopropyl) carbodiimide (EDC) and N-hydroxysuccinimide (NHS), stirring, adding tyramine, reacting, dialyzing, freeze-drying, dissolving in phosphate buffered saline (PBS) solution of graphene oxide, adding horseradish peroxidase (HRP) and hydrogen peroxide, quickly stirring, allowing to stand at 20-40 degrees C, and reacting for 30 minutes to 1 hour. USE - The hydrogel is useful for controlling drug release. ADVANTAGE - The method is simple and easy; and has mild reaction conditions. The hydrogel has high mechanical properties and biocompatibility; controls drug release; and has different drug release behavior based on different pH including fast release at high pH and slow release at low pH. DETAILED DESCRIPTION - Preparing hyaluronic acid/graphene oxide nanocomposite hydrogel comprises (a) preparing tyramine grafted hyaluronic acid by adding hyaluronic acid into 2-(N-morpholino)ethanesulfonic acid buffer solution, mixing uniformly, adding ethyl(dimethylaminopropyl) carbodiimide (EDC) and N-hydroxysuccinimide (NHS), stirring for 30 minutes to 1 hour, adding tyramine, reacting at 20-60 degrees C for 10-24 hours, dialyzing for 2-6 days, and freeze-drying, and (b) dissolving in phosphate buffered saline (PBS) solution of graphene oxide to obtain a mixed solution, adding horseradish peroxidase (HRP) and hydrogen peroxide, quickly stirring, allowing to stand at 20-40 degrees C, and reacting for 30 minutes to 1 hour. An INDEPENDENT CLAIM is also included for hyaluronic acid/graphene oxide nanocomposite hydrogel medicinal composition, comprising a drug including rhodamine B, paclitaxel and lysozyme.
