NOVELTY - System 220 comprises chamber having first liquid medium separated from second liquid medium by barrier/layer having first and second side. The barrier/layer comprises nanopore 200, and nanopore contains first opening on first side 201 of barrier/layer in contact with first liquid medium, second opening on second side 202 in contact with the second liquid medium, an aperture 203 extending through the first and second sides, and a polymerase to the first liquid medium. The polymerase comprises a tether 210. The tether contains a head region 211, a tail region 212, and an elongated body 213 arranged between. The head region is anchored to polymerase and the elongated body comprises a reporter region 214 movable in the aperture. A capture moiety is attached to the tail region of tether, and retains tether in the nanopore. The system comprises a measurement device that measures a first current or flux through aperture or a first optical signal provided by the reporter region. USE - System for detecting event using tethers anchored to or adjacent to nanopores.
