▎ 摘 要
NOVELTY - Preparing multifunctional graphene carrier network, comprises (a) etching by adopting the copper foil covered by the graphene film of chemical vapor deposition growth as electron microscope carrier net, obtaining graphene carrier net and providing the electron microscope carrier with a hole array; (b) carrying out oxygen plasma treatment to the graphene carrier network; (c) adopting 2-(N-morpholino) ethanesulfonic acid buffer solution to process graphene carrier net, where the 2-(N-morpholino)ethanesulfonic acid buffer contains 1-ethyl-3-(3-dimethyl-aminopropyl) carbodiimide hydrochloride and N-hydroxy-sulfosuccinimide salt; (d) adopting nitrilotriacetic acid and biotin activation graphene carrier net after processing; and (e) reacting the treated graphene carrier network with a nickel salt. USE - Preparing multifunctional graphene carrier network used in biological freezing electron microscope. ADVANTAGE - The MFG carrier network can specifically anchor the target biomolecule on the surface to avoid the interface problem of air and water, but also can control the particle orientation distribution, through the related protein purification tag is constructed on different directions of each biological macromolecule, providing a rich Ougui angle, and the structure analysis of the biological macro-molecule is more reliable and more efficient, thus promoting the generality and repeatability of the application of the cryoelectron microscope in structural biology analysis.
