NOVELTY - An immobilized enzyme carrier material comprises sheet-layer graphene-wrapped nano-flower-shaped magnetic nanospheres. USE - Immobilized enzyme carrier material is used for preparing immobilized carbonic anhydrase (claimed). ADVANTAGE - The nontoxic and harmless immobilized enzyme carrier material has high specific surface area, and excellent enzyme binding capacity and binding stability, maintains spatial conformation of enzyme molecules, and economically produces immobilized carbonic anhydrase with high enzyme activity and stability. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for the following: (1) preparation of the immobilized enzyme carrier material, which involves uniformly mixing iron(II,III) oxide magnetic nanoparticles with a solution (A), then adding a solution (B), reacting, magnetically separating obtained product, washing, obtaining titanium dioxide-coated iron(II,III) oxide magnetic nanospheres with a core-shell structure, uniformly dispersing the titanium dioxide-coated iron(II,III) oxide magnetic nanospheres in a solution (C), then adding sheet-layer graphene oxide dispersion liquid, reacting, magnetically separating obtained product, and washing. The solution (A) is a mixed solution of an organic solvent (O1) and an alkaline solution (S1). The solution (B) is a mixed solution of tetrabutyl titanate and an organic solvent (O2). The solution (C) is a mixed solution of deionized water, an organic solvent (O3) and an alkaline solution (S3). The alkaline solution (S1) and alkaline solution (S3) are an aqueous solution of ammonia, an aqueous solution of potassium hydroxide, an aqueous solution of sodium hydroxide, and/or an aqueous solution of calcium hydroxide. The organic solvent (O1), organic solvent (O2) and organic solvent (O3) are aldehyde, alcohol and/or ketone; (2) an immobilized carbonic anhydrase, which is prepared by supporting carbonic anhydrase on the immobilized enzyme carrier material; and (3) preparation of the immobilized carbonic anhydrase, which involves uniformly mixing the immobilized enzyme carrier material with a solution (D), then adding to-be-immobilized carbonic anhydrase solution for immobilization and binding, magnetically separating obtained product, and washing. The solution (D) is a zinc-containing buffer formed by deionized water, a zinc-containing solution and a phosphate buffer. The zinc-containing solution is an aqueous solution of zinc salt. The zinc salt is zinc sulfate, zinc hydrochloride and/or zinc nitrate. The volume ratio of zinc-containing solution and phosphate buffer in the solution (D) is 0.1-1:1. The volume ratio of deionized water and phosphate buffer in the solution (D) is 1:0.1-2. The concentration of zinc-containing solution is 1-100 mmol/L. The pH interval of phosphate buffer is 6-9.
