▎ 摘 要
NOVELTY - Capturing and photoreleasing extracellular vesicles from test sample suspected of containing target extracellular vesicles involves contacting test sample with multiple immunomagnetic particles in test container to create first assay mixture. The immunomagnetic particles each comprises central magnetic particle, targeting moiety and photocleavable linker. The photocleavable linker connects magnetic particle and targeting moiety. The target extracellular vesicles, if present, are bound by targeting moiety to create immunomagnetic complex. The magnetically immobilizing immunomagnetic complexes at first location in container. The first assay mixture is washed, and resuspended magnetically immobilized immunomagnetic complexes in solution to create second assay mixture. The immunomagnetic complexes are exposed to activating radiation to cleave photocleavable linker. USE - Method for capturing and photoreleasing extracellular vesicles from test sample suspected of containing target extracellular vesicles. ADVANTAGE - The method enables to improve activation rate, specificity, stability and accuracy, and enhances immunogenic potential and diagnostic specificity and sensitivity. DETAILED DESCRIPTION - Capturing and photoreleasing extracellular vesicles from test sample suspected of containing target extracellular vesicles involves contacting test sample with multiple immunomagnetic particles in test container to create first assay mixture. The target extracellular vesicles are released from immunomagnetic complex to yield released extracellular vesicles and released immunomagnetic particles. The released immunomagnetic particles are magnetically immobilized at second location in container. The released target extracellular vesicles remain suspended in second assay mixture. The released target extracellular vesicles is isolated from second assay mixture, and separately recovered released immunomagnetic particles. INDEPENDENT CLAIMS are included for the following: 1. a composition, which comprises multiple released target extracellular vesicles, where released target extracellular vesicles each comprises surface modified with of targeting moieties, targeting moieties remaining bound to target extracellular vesicles after release, where target extracellular vesicles remain intact after release, and targeting moieties are active agents. 2. a method for treating or diagnosing, which involves: a. administering to subject composition.
