NOVELTY - Preparing graphene oxide immobilized alkali protease, comprises taking graphite oxide as raw material, dissolving by phosphate buffer solution, magnetically stirring, ultrasonically processing, standing overnight, obtaining graphene oxide solution; and adding glutaraldehyde into graphene oxide solution, cross-linking, centrifuging, and adding alkali protease. USE - The method is used for preparing graphene oxide immobilized alkali protease (claimed). ADVANTAGE - The method above is simple and low cost. The method provides graphene oxide immobilized alkali protease of uniform shape, large amount of active gene on the surface, good dispersity, large surface area and 70.1% average enzyme activity recycling rate. The affinity of immobilized enzyme on substrate is improved. DETAILED DESCRIPTION - Preparing graphene oxide immobilized alkali protease, comprises (a) taking graphite oxide as raw material, dissolving by phosphate buffer solution, magnetically stirring, ultrasonically processing, standing overnight, obtaining graphene oxide solution; and (b) adding glutaraldehyde into graphene oxide solution with pH of 7.0-8.0, cross-linking at 4-37 degrees C for 1-12 hours, centrifuging, adding 100-1000 mu l alkali protease, absorbing at 4-37 degrees C for 0.5-8 hours, centrifuging, rinsing by phosphate buffer solution, and obtaining graphene oxide immobilized alkali protease; the dosage of glutaraldehyde is 0.1-4% of total volume.
