NOVELTY - Detecting target antigen using graphene oxide (10) fluorescence degradation by polymerization of enzyme substrate, comprises (a) forming graphene oxide layer on a substrate, (b) immobilizing capture antibody (11) on the graphene oxide layer, (c) binding target antigen (12) to immobilized capture antibody, (d) binding enzyme-containing detection antibody (13) to bound target antigen, (e) inducing enzyme substrate polymerization by adding enzyme substrate to bound enzyme-containing detection antibody and (f) measuring fluorescence intensity of graphene oxide. USE - The method is useful for detecting target antigen by using the biosensor (all claimed). ADVANTAGE - The method avoids the use of fluorescent dyes, is simple and economical, and directly detects target substance. The biosensor has improved detection limit. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for a biosensor comprising graphene oxide layer, capture antibody immobilized on graphene oxide layer, target antigen bound to capture antibody, enzyme-containing detection antibody bound to target antigen, and enzyme substrate bound to enzyme of enzyme-containing detection antibody, where the target antigen is detected by measuring the decrease in fluorescence intensity of graphene oxide by the polymerization of enzyme substrate.
