▎ 摘 要
NOVELTY - Highly stable co-administration system comprises combined drug delivery system uses graphene oxide as a vector, supports anticancer drugs and apoptotic peptides on graphene oxide, and coating bovine serum albumin with graphene oxide. USE - The co-administration system is useful for cancer treatment (claimed). ADVANTAGE - The system cooperates with anticancer drugs, has strong targeting effect on cancer cells, low side effects on normal cells, good stability, dispersibility and biocompatibility, and disperses stably in water for more than 8 days. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for constructing co-administration system, comprising (i) adding graphene oxide into deionized water to obtain a graphene oxide dispersion, adjusting pH to 9-10, adding 3-mercaptopropyltrimethoxysilane into the graphene oxide dispersion to react at room temperature to obtain a fluorenyl-modified graphene oxide-SH, carrying out centrifugal drying, dispersing graphene oxide-SH dispersed into methanol to obtain graphene oxide-SH dispersion, adding S-(2-aminoethylthio)-2-thiopyridine hydrochloride, stirring to react at room temperature, and centrifuging mixture to obtain graphene oxide graphene oxide- disulfide-amine with a disulfide bond; (ii) dispersing graphene oxide- disulfide -amine in methanol to obtain graphene oxide-disulfide-amine dispersion, adding brominated propyne and triethylamine at room temperature, stirring to react and connecting the alkynyl substituted amino group to a disulfide bond to obtain alkynyl modified disulfide bond graphene oxide; (iii) dispersing alkynyl modified disulfide bond graphene oxide in methanol, adding apoptotic peptides N3-KLA, copper sulfate hepta hydrate and sodium ascorbate having azide groups at the end sequentially, reacting out under a nitrogen atmosphere to pass the apoptotic peptide through the disulfide bond and connecting to the surface or edge of graphene oxide to obtaining a graphitized peptide-loaded graphene oxide graphene oxide-disulfide-KLA; (iv) dispersing GO-SS-KLA and the anticancer drug in deionized water at a mass ratio of 1:0.25-0.5 at room temperature, stirring and dialyzing to obtain a vector complex dispersion carrying the anticancer drug and the apoptosis peptide; and (v) dispersing vector complex dispersion in deionized water, adding bovine serum albumin solution drop wise, and stirring at room temperature overnight.
