NOVELTY - Preparing solid-phase micro-extraction head involves using single-stranded DNA-modified graphene oxide coating, conducting acetone/acid/alkali washings, high temperature quartz fiber coating, conducting silanization using silanization reagent with unsaturated double bond, adding functional monomer, crosslinking agent and initiator in polymerization solvent, mixing to obtain porous polymer solution, taking into test tube, inserting silylated polymer coating silica fiber, removing oxygen, sealing test tube, copolymerizing, heat polymerizing, taking out quartz fiber in drying box, and aging. USE - For preparation of solid-phase micro-extraction head (claimed), which is suitable for tracing amount of alkaloid, antibiotic or nucleic acid molecule substance in the complex biological sample. ADVANTAGE - The method overcomes the problems of conventional coating such as low solid load rate and instability and obviously improves solid load, and enhances solid-phase micro-extraction (SPME) enrichment efficiency. The method provides good reliability, has high recovery rate and so on, has short processing time, has simple operation, reduces labor intensity, avoid use of large amount of organic solvent, is efficient, fast and simple, and avoids secondary pollution of sample pre-processing technology. DETAILED DESCRIPTION - Preparation of solid-phase micro-extraction head involves: using single-stranded DNA-modified graphene oxide coating, carrying out acetone washing, acid washing, alkali washing, high temperature quartz fiber coating, then carrying out silanization processing using silanization reagent having an unsaturated double bond on the surface of fiber, adding functional monomer, crosslinking agent and initiator in the polymerization solvent, fully mixing to obtain porous polymer solution, taking a certain amount of the porous polymer solution into a test tube, and inserting silylated porous polymer coating silica fiber, removing oxygen, sealing test tube, initiating copolymerization in water bath, conducting heat polymerization for a certain time, taking out quartz fiber in a drying box, aging for a certain time at a certain temperature, repeating the steps on the same quartz fiber for several times, conducting silylation process using amino-group containing silane reagent, mixing porous quartz fiber coating with mixed liquid of PBS buffer solution, EDC catalyst solution, NHS solution and oxidized graphite alkene aqueous solution, inserting silane into porous polymer coating quartz fiber, reacting at room temperature for a certain time, taking out the fiber, in the drying box aging under certain temperature, repeating the steps on the same quartz fiber for several times to obtain the oxide of graphene/porous polymer coating quartz fiber, inserting into the mixed solution of PBS buffer solution, EDC solution, NHS solution and nucleic acid aptamer solution, reacting under room temperature for a certain time, then taking out, washing with PBS buffer solution, and drying.
