NOVELTY - Composite hydrogel with immune regulation function comprises nucleus pulposus acellular matrix, graphene oxide connected to the nucleus pulposus acellular matrix by chemical cross-linking and hyaluronic acid connected to the graphene oxide by bridging. USE - The composite hydrogel is useful: in construction of intervertebral disc tissue engineering materials; in construction of composite functional material for regulating immune regulation of intervertebral disc and relieving oxidative stress (all claimed); and for regulating internal inflammation of intervertebral disc and relieving oxidative stress. ADVANTAGE - The composite hydrogel: can simulate natural nucleus pulposus tissue; and has high mechanical property and excellent stability. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for preparing composite hydrogel with immune regulation function, comprising (i) dispersing graphene oxide (GO) in water, adding 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDC) and N-hydroxysulfosuccinimide (NHS) and adjusting the pH value to 5.5-5.8 using alkali liquid, then adding adipic acid dihydrazide (ADH) and stirring at room temperature for 24 hours to obtain GO-ADH, (ii) dissolving hyaluronic acid (HA) in water, adding EDC and NHS and adjusting the pH value to 5.5-5.8 using alkali liquid, then adding GO-ADH obtained in step (i) and stirring at room temperature for 24-28 hours to obtain GO-HA, and (iii) dissolving GO-HA obtained in step (ii) in acetic acid, mixing with GO-HA and 2,4-dinitrophenol (DNP) in a volume ratio of 1:10-30, ultrasonically processing to obtain HA-GO-DNP hydrogel, performing freeze-drying, acid treatment and digestion treatment on the obtained HA-GO-DNP hydrogel to obtain HA-GO-DNP composite hydrogel.
