NOVELTY - Device (100) comprises a substrate (105) coupled to a metal oxide layer (110); a graphene layer (115) coupled to the metal oxide layer; a chemical or biochemical linker (120) functionalized with the graphene layer; and a multiple of Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) receptors that are bound to the graphene layer via the chemical or biochemical linker, where the multiple of SARS-CoV-2 receptors comprise SARS-CoV-2 spike antibodies or SARS-CoV-2 spike proteins, where the graphene layer is configured to have a first phononic energy, when the multiple of SARS-CoV-2 receptors are unattached to target molecules, and a second phononic energy, when the multiple of SARS-CoV-2 receptors are attached to target molecules. USE - Device and method for making and using the devices for testing the Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus. ADVANTAGE - The device has a detection limit of 1 fg/ml SARS-CoV-2 antigen spike protein. The device beneficially has selectivity for the type of protein present in a test sample resulting from the precision of antibody-antigen binding, and the device provide rapid, early and cost-effective detection and diagnosis. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for a method for making the device, which involves transferring a composite film having a Poly (methyl methacrylate) (PMMA) layer and a graphene layer onto the metal oxide coupled to the substrate resulting in a first composite substrate having a PMMA layer, a graphene layer, and a metal oxide layer; immersing the first composite substrate in a solvent and removing the PMMA layer resulting in a second composite substrate having a graphene layer and a metal oxide layer; functionalizing the graphene layer of the second composite substrate with a chemical or biochemical linker; and introducing a solvent containing the multiple of SARS-CoV-2 receptors to a surface of the functionalized graphene layer and binding the multiple of SARS-CoV-2 receptors to the chemical or biochemical linker.
