▎ 摘  要

NOVELTY - Carrier comprises graphene oxide-chitosan-Met-Pro-Gly (GO-CS-MPG). USE - The carrier is useful carrying drug or gene treatment of tumor, and preparing antitumor drugs (claimed). ADVANTAGE - The carrier has small cell toxicity and high biological safety. The method produces graphene oxide-chitosan complex, which improves the dispersibility, stability and biocompatibility of the oxidized graphene; and graphene oxide-chitosan graft cell penetrated peptide, which improves the drug carrying ability of carrier or gene transmembrane efficiency (claimed). DETAILED DESCRIPTION - INDEPENDENT CLAIMS are also included for: (1) preparing carrier comprising (i) adding crystalline flake graphite and potassium permanganate mixture (mass ratio is 1:3, 1:10) solid mixing uniformly, placing the mixture in the beaker, placing in the constant temperature water bath (35-42 degrees C), stirring and slowly adding concentrated sulfuric acid and concentrated phosphoric acid mixed acid (volume ratio is 5:1-10:1), and ultrasonically emulsifying for 30-60 minutes, reacting for 12-72 hours at 50-100 degrees C, finishing the reaction, cooling the product in the air naturally at room temperature, adding product with 500-1000 ml ice water (0-4 degrees C), and slowly dropping hydrogen peroxide till no bubble generated under stirring, centrifuging the mixture obtained (1000 revolutions/minute, 5 minutes), resulting solid was extracted with diluted hydrochloric acid, washing for 2-10 times, washing with distilled water neutrality to obtained solid, adding 20-50 ml deionized water, dispersing into freezing dryer at -10 to -10 degrees C, freezing and drying to obtain the graphene oxide powder, (ii) weighing 10-1000 mg graphene oxide powder, 10-1000 mg 4-dimethylamino pyridine (DMAP), 10-6000 mg chitosan and 1-100 ml dimethyl sulfoxide, mixing, dispersing for 0.5-2 hours by ultrasonic, mixing uniformly, adding 0.1-10 g 1 -(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride(EDC), heating at 50-100 degrees C for 12-72 hours, finishing the reaction, washing the obtained mixture by de-ionized water for 2-10 times, washing with dilute hydrochloric acid for 2-10 times at last using de-ionized water to washing it to be neutral, freezing and drying (-10 to -50 centigrade), grapheme oxide-chitosan complex (GO-CS), (iio) taking 10-1000 mg graphene oxide-chitosan complex with double distilled water for 2-10 times, purifying, adding 10-3000 mg MPG polypeptide, ultrasonically dispersing for 0.5-2 hours, dialyzing and purifying for 2-5 times without water to wash for 2-10 times, removing completely residual unreacted MPG polypeptide, freezing and drying (-10 to -50 degrees C), and obtaining final product; and (2) use of GO-CS-MPG carrier carrying tumor suppressor gene In the treatment of cancer.
